Pharmacological Characteristics

Pharmacodynamic Characteristics:

Ketoprofen is a non-steroidal anti-inflammatory drug. In addition to its anti-inflammatory effect, it also has antipyretic and analgesic effects. Our understanding of the complete range of patterns of action is limited. The pharmacological mechanism of action of Ketoprofen is based on its partial inhibition of prostaglandin and leukotrin synthesis by acting on cyclo-oxygenase and lipoxygenase. Ketoprofen also suppresses bradykinin formation, and platelet aggregation stabilizes the cell membranes of lysosomes, which prevents the release of lysosomal enzymes that moderate tissue destruction.

Pharmacokinetic Characteristics

After intravenous injection into horses, the half-life is approximately 1 hour, the volume of distribution is about 0.17 l/kg, and clearance is about 0.3 l/kg.After intramuscular injection into cattle and pigs, Ketoprofen is rapidly absorbed, with a maximum plasma concentration of approximately 11 microgram/ml achieved within ½ to 1 hour. The average absorption time is approximately 1 hour, and the plasma half-life is 2 – 2 ½ hours. Bioavailability after intramuscular injection is 90 – 100% in cattle and pigs. In case of recurrent infection at 24-hour intervals, Ketoprofen displays constant and linear kinetics, with the above parameters remaining unchanged. Approximately 95% of Ketoprofen is bound to plasma proteins. Ketoprofen is metabolized primarily by the reduction of the ketone group to a major metabolite. Ketoprofen is quickly eliminated from the body, with around 80% being excreted within 12 hours of application. 90% of elimination occurs through the kidneys, especially in the metabolized form.
